![]() | Cloning Method - Will this work? Probably Saturday I will be trying to take a clone from some of the mushrooms fruiting in my B+ grow. First, a quick question. There's one mushroom that's growing pretty big, and is in a cluster. There's another mushroom that's growing pretty big and is not in a cluster. I'm wondering if I make isolate cultures of both of these, if the one will have a propensity for clusters and the other won't. My clone plan is such: I'm going to use the taller half pint jars, with 1 tspn light malt extract to 125 mL spring water. In my glovebox, I'm going to open the jar when I harvest the mushroom. With a clean razor I will lightly skin the outside of the mushroom, then with a second razor blade, slice the mushroom stem open. With a small-bladed box cutter, like this: http://www.amazon.com/60-SMALL-BOX-C...870892&sr=8-28 I will cut a small piece from the inside of the fruit, near the base, and put it in the LC. Does this seem pretty reasonable? I was thinking it might be better to take a second small piece and see how my sterile practice is in a PF jar. |

![]() | I've never cloned before, but have been researching alot in preparation... The plan i had in my mind was pretty similar to urs, except after cutting out the chunk, dip it in dilute bleach solution, and then into the malt jar. Your way SHOULD work tho.. as long as when you cut into the mushroom it doesnt push contams from the outside of the shroom into the sterile insides. I would do multiple clone jars in the same operation... an LC jar costs nothing to prepare, and you increase success rates drastically with each additional jar. As for weather a clone from cluster will create clusters and a clone from non cluster will form non clusters.... I think it has to do more with where the shroom sprouted from and less to do with the genetics. genetics definately would play a roll tho.. cluster is better! |

![]() | The choice of which shroom to use is really up to you. Both of them will have the same genetics, so either should work. Here's a clone TEK I did, this is with an oyster mushroom and I used agar instead of an LC. http://forums.mycotopia.net/oyster-m...clone-tek.html (Simple Oyster Clone Tek) Now I wouldn't dip it into bleach but that's just an opinion. And as you read that TEK you'll see it's better to tear the shroom in half rather than cut it. This will help keep the chance of contams down. Good luck

![]() ![]() | Tear the fruitbody Don't cut it - you'll push external contams into the sterile interior tissue The characteristics of the fruits you clone should carry forward. Make a 10% solution of H2O2/ distilled water 10g hydrogen peroxide 90g distilled water dip your sample in that - I let mine sit for around 10 min. or so I've had poor results with bleach solutions It seems to kill the tissue most of the time |
