Liquid culture question

How other than real cloudy culture how can you tell when your getting good growth? I am using a potato based with Karo Tek so its a little cloudy anyway.

It's a bit of a crap shoot but generally speaking, the degree and tint of the cloudyness should remain fairly constant ,although some particle clumping/sinking will occur..
The myc will just appear as whitish strands/clumps, which sometimes tend to float or sink..
A clean culture cannot be guaranteed without testing in a dry media ,like cake/grain or agar ..So i wouldn't knock up a whole mess of jars without a test run first...

exactly like golly said; knock a jar and incubate. after i see growth ill wait a couple days to see if there is any extra colored growth etc...when i feel im in the clear, i start using the lc. nothing sucks worse than realizing you just fucked a whole bunch of jars, but it gets even worse...wasted hours of mycological labor!

my prob with testing an lc like that before using it... is that u could introduce a contam when you test it...

hope im not hijackin, but what kinda LC lids are you usin that allows you to take out test samples like that without fear of contam?

I plan to use 2 silicon ports and an airport syringe to releave the pressure after sterilizing..

i trust my sterile technique to leave me with about 5% contamination at most. thats a less than 5% chance that my lc is contaminated. if the first test comes up +, i do a second one. if it comes up + in the second one, chances are the lc has got the germ.
ps. i dont even innoculate in a glovebox or nothin. on top of my stove after i sprayed some lysol is all

This will work perfectly. Just sterilize your needle before putting it in the LC jar via a silicone port. Make sure to have the airport in as well (sterilize this needle as well).

after you've taken out the syringe, there is that hole that is left tho that is a possible issue.. even tho the silicon is supposed to self heal... theres no way it can be 100%. right?

I can't say, as you put it, that it is 100%, but I can tell you that it will work for what you are trying to do.

Thanks to everyone. I looked this morning and there is tons and tons of growth. I sterilized the whole batch first off. I used ear plugs on two jars. one jar has 5 in it the other has two in is and some RTV over a hole for an extra sealing port. The last jar I used RTV then two breathing sites with tyvek and med tape it does not look as good I'm going to seal it tonight in my glove box and use ear plugs. I will take pic's so you can see. This Tek comes from (coastalite) It's so far beyond my expectations. I will get some berries ready and test the LQ I am going to use the one I'm concerned about first though as well as the others O yeah I also put cling wrap around all the jars just for a little extra precaution.

No words now for how excited I am. but I have been here before and crashed & burned so please say a prayer and send it up with some

potato-it doesnt leave a hole, hence the term self-healing. ive used these ports several times a piece and never had a problem. in fact ive never thrown one out yet which means somewhere im still using the first one i ever made.and they take abuse-figure 500cc per quart is the average amount i put in, slurp it out 10-12 cc at a time so about 50 penetrations(ha ha) per lc. works for me. just make sure you use rtv to make the port

and I wish I was a better camera man. I inoculated these Monday around 7:30 am. I used extra ear plugs to have more places to draw from. I just worry about using the same port over and over thats all. The single jar is the one I switched to the ear plugs. You can kind of see the spot on the lid.

I'm seeing lots more growth.I am just thankful it's here. I do think it could be made lighter if you filtered it more.The tek called for the skins to be left on which contributes to the color. It could be I left it on to long but it is my first go at LC so I'm pleased. If you have any experience in how the lc will shorten time I really appreciate it. I still have yet to get everything to go right. I am close on some rye berries again and need a good sub. I might give straw a go not sure yet so all suggestions welcome.
