SWIM just cloned to cardboard for the first time. He is planning on making a few LC's out of the piece. He is wondering if it would be ok to dip each piece in a light solution of H202 before putting each one into a LC jar? Could the myc. handle it? |

I'm not the best person to answer your ? , but i feel a dip in a light solution should be ok , let's see what others say. I am very interested in seeing where you go with this Smiling Turtle, as i have recently begun using cardboard myself. Lots of fast growing, healthy white mycelium on this end. Please post more!

well this FOAF of mine is having troubles keeping the cardboard moist throughout the colonazaation process. Only part of his piece is colonized and he wants to get it in a LC or something before it totally runs out of moisture. SWIM needs to get his cheap ass some petri dishes |

Hydrogen peroxide is safe for live mycelium since they make peroxidase. Spores can't handle it. A H2O2 dip has worked well for my cloning to LC. Good Luck and let us know how it turns out.

Try placing it in a zip lock bag. It will keep the moisture in the bag until it is colonized. As far as a solution of H2O2.... I do not think it will help you any. That's just my opinion. I can not say for sure that it is a waste of time. If cloning is the way you want to go I can only suggest working with agar. I tried to clone off of cardboard and had mixed results. I was able to get the myc to grow, but had issue transferring to LC. It seems I would always end up with a contaminant. BTW... baby food jars work well for agar. Much cheaper than glass petri's and the agar can be pc'd in the jar to be assured that it is sterile before inoculation. this saves the step of pouring hot agar in a cumbersome glovebox. good luck. ggod |

I've been playing with a bit of card board as of the late too.Although I've been just trying to isolate rhizomorphic strains in open air instead of transferring to grain or lc.I would think it would work assuming the H202 kills the contams off.You may have to transfer the mycelium to new sterile cardboard and select clean mycelium to then in turn move to a liquid culture.It all depends on how sterile your cloning was done and how efficient H202 is at cleaning.I've only done a little with h202 so IDK on that one but what I can tell is h202 can strongly weaken the mycelium so keep the dip brief.
