Alright, so a few weeks ago I tried making some myc syringes by injecting sterile water into a fully colonized PF jar, shaking, and then sucking the myc water back into the syringe. Made two 10cc syringes and inoculated 8 PF jars. Had 4 jars left over so they were just stored on the shelf for later use. After the 8 jars had been incubating for 2 weeks and still showed no growth, I figured the syringes were bad so took the jars out to free up space for some bulk subs. Those jars were just sitting on a shelf for the last three weeks, and while rummaging around up there today, behold! Growth in all the jars. They were quickly placed back in the incubator. But now for the weird contam, one of the 4 jars I never knocked up was completely filled with what looks like trich to me. It's weird because the jars were PC'd for 70 minutes at 15 psi. The tinfoil was never removed from the jar after cooking. Anyone else have something like this ever happen? Hope it's not some bizarre mutated super trich that's immune to pressure cooking! I believe it was prolly stray airborne spores that entered the jar during the cooling down time. Since your lid is inverted there was not an air tight seal. Your nocc'ed jars had a good head start because of the LC. and any stray contam spores were beat out.

I cant see too many spores making it through a pc run.I usually pc my jars for 90 min @ 15 psi but 70 should've been enough.Hope you get it all figured out.GLUK.

I Go a little overboard with sterility i guess. I put the still presurized pc in a steriled tub to cool down. The jars that are not air tight breath due to temp changes in the area there in. It only takes a few spores to cause trouble. You had good success other than that. I did'nt see any green in the jar but it may have not thrown spores yet. Hard to say what the growth is.

The picture doesn't really show the colour, but there's a lot of green in the jar. I'm thinking that this may just have been an unusual isolated case. This is the only contam I've had out of 36 jars, I'm also kinda anal about sterility, it's key to good success. Moose |
