Liquid culture questions

Hey everyone Ive got a couple questions would appreciate a little help

1. I plan on making 10 liquid cultures with 4 grams of honey and 100 mls of water. What would be the best way of sterilizing them?

2. I plan on innoculating each with 1 cc of multispore. Obviously when I see mycellium I can suck it up but how long will I most likely have to wait to have a decent amount of LC in each jar?

3. What room temperature should I store the liquid cultures at?

4. Should I pay any concern to light or air (on a shelf vs in a drawer)?

5. If I had to transport the LCs by driving for a couple hours what would be the best way? Im thinking place each jar (which has foil on the lid and micropore tape covering the injection hole) in a ziplock bag and then place them in a cooler, and then secure them within the cooler and secure the cooler so that they arent bumping around and spilling in the drive.

In the PC @ 5 PSI for 25 minutes is enough.

7 to 14 days should give you a decent amount of growth

Are we talking long term storage or incubating?

Long term storage you will want to refridigate them so they stall, you can keep them for about 6 months.

If you are asking about incubating temps then 78 to 82 is good

No when growing a liquid culture light isn't really a factor although its nice to keep them is a dark warm place like a incubator

Sounds good but, only leave put the tinfoil on for the drive But, you don't want them splasing around and getting that tape wet, so best off to use pint jars and only fill them half way this should help prevent that, but, as riding down the road can be bumpy sometimes you will want to be gentle. Also, I would put a layer or strofoam down in side the cooler and cut whole in it slight smaller that the jars, make this about 4 inches deep them force the jars down into those and this should keep them nice and secure them cover them with strofoam or something soft that will fill the cooler up the rest of the way and close the lid. This will make sure that the jars are secure and resistant to shock and while help prevent splashing as well.
