FOAF tried his first ever spore print, used glovebox and Hippie's oven technique to sterilize foil squares. Point is, he thinks he picked the cap a bit too early, and after more than 12 hours the print is only visible through the baggie if you hold it at an angle. Is this still something that can be used to make a syringe? There are definitely spores there, it just doesn't seem like it would be enough to do multiple syringes.

I throw out the first print in case it carried any contams with it, and the second one is the keeper for me. Sometimes I can get two. It doesn't matter if it's dark or not--if you can see spores there are plenty there for your use. |

I generally wait until I see the first dusting of spores on the stem before cutting the cap for printing. The cap should be completely opened and flattened out across the top.

instead of starting a new thread i am just curious what everybodys procedures are for there cleaning of all the equipment used for making spore prints. (ie) foil,razor blade,glovebox, or jars,plates, and your storing process folded foil, open foil, and bags there stored in fresh 2-1/2 inch by 4 inch bagies ect. and rubbing everything down with iso. masterplain88 |

I sterilize everything that will fit into the pressure cooker and not melt. My cloning kit is a stainless steel scissors and tweezers as well as a stainless scalpel handle and a stainless probe. I wrap them up in aluminum foil and run them through the PC. Same thing with syringes. When the aluminum foil packets come out, I toss them into a gallon Ziploc to keep dust off the exterior. The packets are only opened in front of the flow hood and the aluminum foil "cover" is opened up to make a sterile work surface (if needed).

I saw in another thread where you describe wrapping foil squares in another layer of foil, bake at... 400? something like that, for an hour, then place the packet in the glovebox, to be opened inside, where the now sterile squares can be safely used.

It sounds cooler than it is! Just a stainless steel dissecting probe with a triangular head. Good for digging out stem tissue and easy to sterilize with a mini-blowtorch lighter. Lots of dissecting tools and dentist tools are good for cloning work.
