i have just started the fraction sterilisation process today and the jars will be ready to inoculated in a couple of days time i have a good idea about what im doing now but am still abit unsure about how to clone the mycelium and mushrooms so that i can isolate them so they can be used in my next grow thanks, ps i am using the mazatapec strain

I dropped mine to agar, let it grow out and made a Karo LC using a recipe from here. Cut tissue and dropped it in the Karo solution inside a glovebox. The recipe for the agar i used was agar flakes from a natural food store, potato flakes and some karo. 2 tbl pot flakes, "not reefer" lol 1/4 cu agar flakes 1 tsp corn syrup 1 cu water. This recipe will make more than you really need. I sorta 1/4'rd it. I dont have petri's, i used small jars with airport in the lids. For the Lc, i used 1 tsp Karo per 100 mls H2o |

Cloning means taking a small amount of sterile tissue from an adult mushroom and growing it out on agar, in LC or on grain.

Once myc is growing on agar, you can move it to any media you desire - agar, grain or LC. You can use a syringe and create a slurry of water/myc to inoculate agar, grain or LC.

I like the classic Potato Dextrose Agar (PDA) from sporeworks. I've used it many, many times.

The Mason jar is pre-mixed and pre-sterilized agar. You put in hot water until it melts and then pour your plates. A Mason jar will make about 20-30 plates. The jars of agar are just powder. You add water, heat and sterilize. Then you pour plates. The jar will make around 7 liters of agar (about 14 Mason jars). The powder is more expensive because it makes a lot more agar.

I would recommend cardboard to clone on. Easy to work with and doesn't contaminate as easily as agar. I've had a bunch of agar plates go green in a couple days. I've only done 3 tries with cardboard but it has never contaminated for me. I had one piece of wet cardboard lying around for almost a month and no contamination. I have to add that I have never sterilized the cardboard.
