![]() | 1st g2g of my career
I will be attempting my first grain to grain transfer of my life tomorrow. I about two quarts of wbs already colonized. Today I preped six quart jars of rye grain via the one step method. The moisture content seems to be uniform after a good shake or two. I will be useing my own homemade glovebox. I need to know a couple of things. Would it be best if I went ahead and sterilized my tools in the PC wrapped in foil or can I just wipe them with rubbing alch? I will need to cut open the mycobag that the wbs is sealed in so I guess I'll need some scissors and a tablespoon. When I get everything into the glove box should I wipe down the jars and the mycobag before attempting the transfer? I would assume so, but hey how should I know. Is there any good advice that some pro could pass on to me before going through this process? I will post any results from this little adventure.

you can spray all your jars with lysol if your worried. if u pull them directly out of the pressure cooker and put them directly into your glovebox you should be fine. if im in doubt. i always either wipe the outside with alc or spray with disinfectant spray.

I agree with eatyualive. I usually do everything requiring a sterile environment in my bathroom that I have previously removed all cloth materials, cleaned and sprayed down with lycol and air sanitizer. I shut off the fan in my A/C unit and put a towel down by the door to keep the air from circulating too much. Give a good oust spary. Wash my hands, or if I'm feeling dirty, I'll wear latex gloves. other than that I follow that eatyualive says exactly.

I sterilize all my tools in foil wrapped packages. Cleaning the glovebox and everything in it isn't a bad idea. The most important thing is to move slowly and precisely. Quick movements stir the air and defeat the purpose of a still air glovebox. I would strongly suggest you do at least one practice run (3 or 4 is even better) before attempting the sterile work. Your chances of success go way up when you are familiar with the steps. The last suggestion would be do at least six of everything. Plan for 50% contamination and you will almost never be disappointed.

update............ I did my transfer on monday and it seems as if the jars of rye grain are colonizing well. I can not see any sign of contams. I did six jars of winter rye and all six are trucking along. I closed myself in the bathroom as suggested. I also sterilized the need utilities. I sprayed oust around very liberally and used alcohol to clean the glovebox as well as the outside of the jars holding the rye and the mycobag of wbs. I spilled the alcohol on my bathroom floor and ruined the wax finish. My girlfriend stood on my skull and hopped when she saw the nasty dull spots(gasp) on the tile floor. Thanks to all who gave advice and I will post pics and results when I get them. I will also pass on the good advice when I can.
