Hi folks, Foaf tells me that he made have made a silly mistake when transfering spores to agar plates. He dipped his noc loop in alcohol but in his haste he didnt let the alcohol dry fully first. Has he f****d it??? First time using plastic noc loops, got confused without using lighter He's fairly new to agar and his glovebox is quite small which caused some discomfort thus distracting him and causing the lapse in concentration. What is the usual length of time it takes to see growth of woodlovers on agar at 24-25 C ?? He wants to know how long to wait before assumming that they are f****d and starting over....

Its been a little while since foaf germed woodlovers on agar.. Foaf thinks it took about 3-4 days with MEA. foaf remembers his azures taking a day or two longer than cyans and subaerangosa to germinate. foaf uses a torch with a metal innoculation loop, so not sure on alcohol spore killing abilities. If anything you will hopefully just get less germinations than none at all. as long as the plates are still clean you could still streak more spores on them. maybe wait a week and then toss if you don't want to streak more spores on the plates. |

Every woodlover I've worked with usually germinated by the 7th day. You may have some exceptionally dry spores but 7 days is quite awhile. Gotta let the alcohol evap first. If the loop was in a sterile package to begin with, no need for alcohol wipe. If you re-use them, like I do sometimes use several at a time. Place them in a shot-glass of alcohol and soak for a min. or two then pull them from the alcohol and allow to air dry. you may have to do this as part of your glovebox prep work so they dry within the glovebox In this fashion you can be using one loop Have a clean one waiting Re-soak the loop you just used Move on to the next culture with a new, dry, sanitized loop an assembly line of sorts - you'll work it out. And Yes first-hand experience has shown me that if you don't allow the loops to dry the alcohol will nuke your spores - end of story

Checked plates this morning and 2 are blatently contamed, have chucked them. Has anyone had success germinating woodlover spores on cardboard?? Am thinking this might be a way to beat contam's. Was guna make up some PC'ed PP5's with card in the bottom, then once colonized, simply add a thin layer of sawdust, then slowly add thin layers of small ash chips as colonization continues. What do folks think?? Cheers

I have used agar that i mixed a few of the wood stove pellets (no additives) and my wood lovers did alright on those. I also tried with cardboard with limited success but didnt try that for long as I was having luck with the agar.
