Best choice for 'rare' spores ?

I've been away from the hobby for a long time and am, as Barack Obama says, 'all fired up and ready to go' again. But first, I need some good advice on how best to work with a new syringe of Penus Envy from one of the sponsors. I know the strain has been problematic in the past but can't resist trying it again.

I have fresh petri / agar plates as well as dextrose jars with stir rods available along with grain jars.

So I'd like to make the most of the syringe while I'm working with more robust strains. In the meantime I'll become familiar with the more recent advancements in the hobby.....from what I see, there have been incredible leaps in knowledge and teks.

Shall I start by putting a few drops on agar ? How much in the dextrose jars and still have some left over for future use ?

agar's a good beginning, get a good isolate to work.
a lc should be good even with 1 cc of sporewater,
it'll grow from there.

Once I start a syringe, I just use the whole thing. Just make sure you've got good sterile technique and as many fail-safe's as possible. When I did my LCs, I did silicone ports and used a glovebox for inoculation. Every jar I've inoculated has looked great.
