![]() ![]() ![]() ![]() ![]() ![]() ![]() ![]() ![]() ![]() ![]() | Here are some pics showing the benefits of strain isolation on agar. From a single spore print, many strains can be isolated. For this experiment, the fastest and most rhizomorphic strain was colonized out, along with the slowest and most cottony strain. All the mushrooms below, came from the same spore print, at the same time. A single zigzagging swipe on agar from an innoculation loop. This should show you how much variation is possible when just using multispore innoculation. One half of the pie plate was filled with one strain, and the other half filled with the other. This is the best way to test your strains and see very clearly which is most favorable on a genetic level. The next step in the process would be to then clone the largest, fastest fruitbody on the "good" side of the pie plate, and you then have your super strain of Cubensis or whatever species you are working with. These happen to be Treasure Coast. Quote:As you can see...the half that's on the right in the pictures is obviously superior to the half on the left of the pie plate. That is the side that the cultivator should clone and use for subsequent large trays |

Cool thanks hip nice workIts amazing some of the non mycological subjects you can learn about from this site. I always knew inbreeding was bad but didnt know why. I have a question tho. If you spawned say with 10 multispore cakes and cloned the fastest largest fruit cluster would this have a similar affect of isolating a strong substrain? Or is the disadvantage to cloning fruits from multispore that u cant observe the speed or vigor of the mycelium growth till you recolonize with it ?

shrooms actually have a safegaurd against inbreeding so it is nothing to fear. however, it is possible to slowly isolate or breed yourself into a non-fruiting strain< "substrain", but it is quite rare. when choosing prints try to take them from the clustered areas whenever possible . once cloned or isolated you will not have to worry about it quite as much.

well.. i am at work and don't have all the resouces at my finger tips. sorry. but, not all the spores from the same fruit can mate with each other. chances are about 1 in 5 (rough estimate) that hyphae can mate. then the mating types have different genetic codes. the dna is exchanged causing the monokaryotic myc (single cell nucleat) to become dykaryon (bi-nucleat). this is also how the proper term of "strains" are made. i'll try to find some links if i get a chance

No need- that's about what I thought about the situation. Mating types reduce, but don't eliminate inbreeding. I wondered if there was more to it than that. (edit- but more info is always good... if you find links, please post!) Armadillos have a cool genetic defense- a litter of armadillo pups is always identical quadruplets. Four males or four females, no possibility of inbreeding. |
