tek for syringe making? [merged]

Hey all.

I know many make syringes by using a glass, scalpel and syringe. Most will put the water in a glass, scrape the spores into the water, stir it up and then suck up the spores in a syringe. When you do this the spores want to float on top of the water and stick together and stick to the sides of the glass.

If you aren't using jetdry, which would help the spores not stick together, I recommend putting the spores in the glass FIRST.

Scrape the spores into the dry glass. Then you can squirt all your water in on top of the spores. It seems to break them apart much more effectively and distribute them throughout the water more evenly, keeping large chunks from sticking to the sides of the glass.

Something I have been doing is with sterile water in dish/bowl/cup, remove the plunger from the syringe, add some spores in syringes, replace plunger and draw water. (All of this done inside a glovebox, of course).

Hip, this brings up a good point. I have a cheap ultrasonic jewelry cleaner, however, there are cleaning chemicals that are put in it. You're saying to just put the bowl of water on top of the cleaner and it will work?

should contact the metal bottom of the unit
so the sound can conduct better

Any hints for using an ultrasonic mister for the job?
I've tried putting jars of water on top of the transducer elements, but it doesn't seem to agitate the water, so I don't think it's doing much of a job.

If you are using good prints that won't happen, the spores will 'disperse naturally'. However, if you are having problems with lousy prints that clump up on you then I suggest letting the spores soak overnight and also just give the water a little jiggle. Not a swirl, which will spin the spores to the outside where they will stick to the glass, but a jiggle that upsets the middle of the surface of water. It's amazing how the spores sink like rocks once the water tension is broken.

Another idea - a shot in the dark - lay your syringe on your cat. I recently read that a cats purr causes almost the same vibrations as ultrosonic that is used for pain. You can actually hold a purring cat and it will help reduce pain. Maybe it will break up those spore clumps too!

Question best tek for syringe making?

after yesterdays contam. I got lucky with having a print sent my way,as well as hoping to hook up with one of hillbillies kits!I just want to know the best tek for going from that print to a syringe,any suggestions are appreciated!

Print in a new plastic baggy.

Leave baggy open for air exchange while printing.

When ready to make syringe.

Simply take new syringe , fill with sterile water , inject water into the baggy , mix with your fingers OUTSIDE the baggy , Suck water back into syringe.

DONE...

I find this ti be the easiest way for me.

If its a big print and very dark, you could get 10.

Be very careful if you decide to print in a baggie. The baggie cannot touch the top of the cap at all. You'll need to wipe down the top of the cap with a damp cloth and insert some tacks or a nail into the top of the cap to prevent the baggie from touching it. Personally, I don't recommend it at all...

If like to put my prints on glass slides. It makes the baggie/syringe method much easier and you can scrape all the spores off the glass if they get stuck. I have never tried it with a print on paper in a baggie. Slides are easy to clean. Just wipe with alcohol and flame it.

I mass print my slides in a large gallon sized ziplock and then transfer the slides to individual snack size ziplock bags for storage. The snack size ziplocks work well with 10cc of H2O in them. Easy to manipulate.

i do something similar to silly.. but personally i don't like printing directly inside a bag b/c any water that gets on that plastic isn't gonna dry quickly.. Which can spread bacteria.. My tek is as fallows..
Materials needed:
tyvek
scissors.. one large.. one small
2 half pint jars
2x3 ziplock bags.. you can get them on ebay for about $5
tweezers
foil
ISO.. paper towels
large tupperware.. i use a 6qt.. about 6x12
thumb tacks
gloves, mask, oust
(optional) shot glass and decedent

ok.. first thing is first.. take your tyvek and large scissors and cut the tyvek in to about 1.5 - 2in squares.. i like to cut about 25 or 30 each time..
now put all of your squares in 1 of the half pint jars.. then put on a double layer of tyvek and the band with foil on top.. no solid lid
then put your small scissors and tweezers in the other half pint jar with the same lid set up
pc those for 20 mins (or whatever) i normally just throw them in with what ever i'm pc'ing at the time.. i know it's a bit extreme but i like to make sure my stuff is sterile
now when it comes time to print..
put on your mask.. so you don't breath bacterial all over everything
gather your stuff on a cleared table
tupperware, foil, 1/2 pint jars from pc, thumbtacks, gloves, oust
wipe off a good working area with ISO..
put on your gloves and clean them with ISO
wipe down the tupperware with ISO.. make sure to get the lid too
then take a piece of foil off the roll and lay it inside the tupperware.. wipe it down as well
also take a minute to wipe down your thumb tacks and set them aside
you want everything good and clean..
now while i go to retrieve the caps i spray a good dose of oust into the room
get your fruiting substrate and set it off to the side a bit
take a quick moment to determine which caps and how many you are going to print.. you can go ahead and put your thumbtacks in the tops of the ones you've chosen
once you got it figured out.. re clean your gloves with ISO
take the lid off both the half pint jars..
take your tweezers and set out a piece of tyvek for each cap you plan on printing.. if they are small caps i put two on one piece of tyvek.. if they are large caps I use 2 pieces of tyvek..
take your small scissors and cut the caps off.. make sure to hold onto the thumb tack while doing this..
now just transfer the caps one by one to their respective tyvek squares
once all the caps are transferred put the lid on the tupperware..
find a good clean dark spot to let it sit for a 12-24 hours
(i put the lid on tight while carrying it, then take it off and leave it slightly ajar once it's sitting where it will stay)
once your caps are done printing
.....
set things up exactly as you did for the first time as most caps can be printed more then one time.. go ahead and lay out the tyvek and put the lid on the tupperware
go get the other tupperware from it's printing spot .. clean your gloves again
right after cleaning your gloves take out your ziplocks and go ahead and crack the seals.. as this will be much harder to do with tweezers in hand later on
now take the lids off of both the tupperwares
then just pick the caps up by they thumbtacks and transfer them to the new tyvek..
take your tweezers and wipe them with ISO.. then flame them with a lighter for a second.. wipe them with ISO again..
now in one hand hold the tweezers .. and in the other hold your ziplock baggie
take the tweezers and use them to open the ziplock.. since you cracked the seal earlier just put them 1/4in inside the bag and move them back and forth along the seal
now pick up your print and place it in the bag
i like to leave the bags cracked open and put them back in the original tupperware with a small amount of decedent for 24 hrs.. but this is entirely optional..
if you chose not to do this go ahead and seal the bags right after you place the prints in them.. if you go the decedent route.. just seal them up with a gloved hand when you remove them after drying..

the reason i adapted this tek was for it's simplicity and ability to do in open air.. it also makes making syringes much quicker and easier, which can also be done in open air with good clean procedure.. which means you can do the same thing with shooting up jars and or myco bags... just be sure to wipe everything down with ISO or bleach and give the air a good shot of oust before starting.. one can actually go through the hole process with no glovebox.. just need a good clean draft free room, gloves, mask, ISO and paper towels..

has anyone here had any experience with ordering syringes and needles from medical supply companies? it seems that you get 100pk for around 35.00 usd. alot better than 10.00 a piece.-- 10cc with 1.5" 18 guage needles

depends on the company, and what state you live in. In some states, it is illeagal to send syringes and needles through the mail. Check with the company first.

Your local feed and seed supplier will have them too. You know the place farmers go for cattle feed horse supplies, and gardening equipment.

haven't tried the cat
yet
but have tried various vibrating objects
[]
and still find ultrasound far superior
in breaking up spore clumps.
after numerous experiments
the only snag i've found with ultrasound
is you must watch the water temperature
so it doesn't go too high,
the sound waves heat the water in passing.

my foaf put the spores in a sterile shot glass then scrape a small amount of spores into it then with a syringe full of sterile water he just puts a few drops on the spores and mix them with the needle till its broken up then he squirts all the water out then suck it up back and forth a few times. maybe he'll get an ultrasonic device someday but until then thats how he does it

Anybody had any problems using a tiny bit of Jet Dry when making syringes? I tried it just recently and the spores don't seem to have any trouble sticking to the syringe or each other. I haven't used them yet, and was wondering if anyone had noticed any negative results from using JD, like decreased viability or anything.

Because if there's no reason not to use it, jet dry apparently works well.

I make my prints in small jars, then suck up PC'd water (jet dry added before PCing) and blast it at the print. With jet dry, nearly all the spores easily came off the glass and mixed into the water without clumping. Without JD, I needed to scrape, there was some clumping, and they stuck to the syringe.

Strapping a syringe to a cat would never have occurred to me, thanks for that intriguing tip. Would a small, yipping crotch-stomping dog work in a pinch? The cats around my place are too hard to catch, but the neighbor's little dog is friendly enough and his yips are short, high-frequency bursts...seriously, I love that kind of thing, especially explaining it to someone else. I don't think I'd like to explain to my neighbor why there's a syringe stuck under his little preciouses collar, however, so tests will have to wait for now.

Aw, hell. I keep them in the fridge, so I hope that prevents it, at least until I shoot 'em into something. Might be a good thing if intending to use them soon after making the syringe...?

Anyway, thanks for the info, I'll keep an eye on 'em.

Syringe Making Question

Well i just attempted to make my first syringe. The cap that I used was maybe 1in in diameter. I scraped the spores into a shot glass, added some water, and drank it up through the syringe, and it looks like theres no spores in the syringe or the shot glass.... I don't understand it b/c their was absolutely no air currnet, i saw the spore in the shot glass b4 I put the water in, and the syringe looks like its just filled with water.

Do the spores get larger as they absorb the water and I should pssible see them in the water more clearly?

Any advice would help.

The spores are probably in there. All the syringes I've ever gotten from vendors were crystal clear.

Conventional wisdom says to wait 24hrs before inoculating jars so the spores can re-hydrate.

Yep, unless you have very large clumps in the syringe, spores are seldom visible.

If you leave it undisturbed for a few months, you might get a very fine "film" of spores settling on the bottom, but usually they're invisible to the naked eye.

Alright, I was just worried because I've seen pictures where the water is like brown from all the spores that are in there. So when I saw nothing I was like oh man I screwed up! Well i'm going to be making two more jars tomarrow to test them out. A BFR and a whole rice, Im making both because I want to try the whole rice, and the BFR is a test to make sure I didn't contam the spores.

Just wanted to update on this, I tested my first ever syringe of Treasure Coast and got 0 contams! Very happy that I got it to work well. Thanks for all the help. I'm not not going to have to buy spores again.
